Did you forget to grow an overnight culture? Don’t want to wait all day for your culture to regrow? No problem! Our protocol will work for fresh liquid cultures, plated colonies, and even bacteria that has been stored at 4ᵒC for up to a week before use.*
No matter which type of bacteria you use, just harvest the cells by a single centrifugation step, resuspend them in the chilled premade buffer, and aliquot into individual tubes: no wash steps required!
Once prepared, the competent cells can be stored at -80ᵒC or used directly to transform your plasmid DNA or ligation products.
Not only is this kit easy to use, you will be amazed by the results. In fact, competent cells made with the GigaBiTe E. coli Transformation Kit are typically more effective at taking up DNA and forming colonies than commercially available cells, with efficiencies of up to 109 cfu/µg, depending on the bacterial strain used.
Competent cells prepared using our product will save you even more time during the DNA transformation process, since the standard incubations, heat shock, and recovery steps are all optional. Instead, add your DNA to the cells, spread the mix directly onto LB plates with ampicillin, and you’re done! It is important to note, however, that transformed cells should be allowed to recover briefly in media without drug when selecting with antibiotics other than ampicillin.

*Although any source will yield transformation-competent cells, best results occur when bacteria is grown in liquid culture at room temperature or below (16-25ᵒC) until it reaches an OD600 of 0.4-0.8.
