The Bradford Protein Assay is a highly optimized, colorimetric assay designed for fast, simple, and accurate quantification of total protein in solution. Based on the established method developed by Bradford (MM, 1976), this kit uses Coomassie Brilliant Blue G-250 (CBB G-250) dye, which binds to proteins and produces a measurable color change.

Under acidic conditions, the dye primarily exists in its doubly protonated red cationic form with an absorbance maximum (Amax) at 470 nm. Upon protein binding, a shift in absorbance from 470 nm to 595 nm occurs, resulting in a color change from brown to blue, which can be easily measured using a spectrophotometer or microplate reader.

The CBB G-250 dye exists in cationic (red), neutral (green), and anionic (blue) forms. When bound to protein under acidic conditions, the dye shifts to the anionic blue form, producing a strong signal at 595 nm.

The dye binds primarily to basic amino acids like arginine and aromatic residues, creating a stable color complex with a constant extinction coefficient over a 10-fold concentration range when complexed with albumin.

This assay follows Beer-Lambert’s Law, enabling precise and consistent quantification by selecting the right dye-to-sample ratio.

• Easy Assay Format

Compatible with both cuvette and microplate formats—just mix a small amount of your protein sample with the reagent, incubate briefly, and measure absorbance at 595 nm.

• Supplied with BSA Standard

This kit includes Bovine Serum Albumin (BSA) as the protein standard. BSA is widely used in protein assays due to its consistent and significantly stronger dye development compared to most proteins, including gamma-globulin.
